Since I'm a long time RHPS fanatic, I thought I'd share my two cents. I did a fan art of Lee as Rocky for my girlfried. She thought it was perfect. She and one of our friends both thought I should draw ones of all of them. Maybe I will one of these days! Anyway...
Frank: Orochimaru ('nuff said.)
Janet: Ino (I'm not sure. Sakura might work, too, but I can't stand her.)
Brad: Kiba (Or not. Brad's tricky.)
Riff-Raff: Kabuto (Agree with you there.)
Magenta: Kurenai (Sexy!)
Columbia: Gaara (I love and cosplay both of them! Short red hair, no eyebrows, heavy on the eye-liner; it just fits!)
Dr. Scott: Hiruzen (Seems the type, but I could be talked out of it.)
Rocky: Rock Lee! (He's got the hair and everything!)
Eddie: Choji (He'd be so cute!)
Criminologist: Jiraiya (It never occured to me, but he is a great choice!)
Perhaps I based this more on looks, but oh well.

I think that Columbia should be sakura... *hides from Sakura bashing fan girls* Dont worry! i dont like her either... but it would be awesome... seeing as Columbia is in love with eddie... (Sakura and Sai =])

You need to put Kakashi in there somewhere as well. Otherwise, your casting is pretty good. Sasuke as Dr. Frankenfurter = pwn.
